The relationship between catalytic current and the concentration
of hydrogen peroxide showed aMichaelis-Menten response.
The apparent Michaelis-Menten constants (KM
app) for the
immobilized proteins,obtained from the Lineweaver-Burk plot
(1/I vs 1/[H2O2]),are reported in Table 2.The values forHRP and
Hbwere lower than those forMb,indicating thatHRP andHb in
the Sclg-borax film exhibit a higher affinity for hydrogen
peroxide.These KM
app values were smaller than those obtained
by Hb/Au-SBA-15 (2.87 mM),48 HRP/Au colloid/Cys/Au
(2.3 mM)49 or other HRP modified electrode (5.5 mM).47 Thus,
it can be concluded that the three heme proteins entrapped in the
Sclg-borax film have a high affinity for H2O2.
The electrocatalytic behavior of heme immobilized in
Sclg-borax PG electrodes in the presence of hydrogen peroxide
was also studied and the main kinetic parameters were evaluated
for comparative purposes.The value of KM
app was found 22mM
for the heme modified electrode,giving a clear evidence for the
sensitivity of the enzymemodified electrode,with a KM
app smaller
than 3.94 mM.
Additional experiments were performed to test the permeability
of the Sclg-borax/proteins film toward hydrogen peroxide.
