是的。of course是一个插入成分，就翻译为“当然。。自然。。。”
不要把course分开来看

可改为
He is of course older than me

此类轶闻的问题当然是你无法区分的。这是表语从句，is前的是主语，that引导表语从句。is不可省，is是句子一部分。of course 是当然的意思，去掉对句子结构无影响，但当然的意思会去掉。
