This aritcle through the steps such as PCR,transformation,distilling plasmid,EcoRI,connecting main links,purification and so on,in order to built the plasmid pET28a-NULP1.First,amplifing the nulp1 through PCR,and connecting the nulp1 to the carrier EcoRI pMD18-T then testing and sequencing,when you got the correct nulp1,then connecting nulp1 to carrier pET-28a,built the regrouping plasmid pET-28a-NULP1 successfully.

本文通过PCR、转化、提质粒、酶切、连主要接、纯化等步骤构建了质粒pET28a-NULP1。先通过PCR扩增出了nulp1，并将nulp1连接入pMD18-T载体酶切检测并测序，得到正确的nulp1，再将nulp1连接到pET-28a载体，成功构建了pET-28a-NULP1重组质粒。
