mustn't be不能用来表达推测的用法.所以你的答案是错的.
而couldn't be是可能不的意思,正好用在这里表示推测.

否定结构是 can't 和could't 表示不可能

在英语口语中很少用mustn’t be，如果需要通常用needn’t表示
在这句话中表示不可能应该用could表示可能性，而不是能够的意思
