这两个否定句的含义完全不一样：
Can't you come to the party?- 你不能来派对吗?（本意还是希望对方来.）
Can you not come to the party?- 你能不能不来派对?（本意是,你别来了）.
这两句都和原句有些不同.看你需要表达什么意思了.

都可以，not可以和be动词、情态动词、助动词缩略至于主语之前，如果没有缩略就只能放在助于之后。
