都用best.
两者比较,用better.三者或三者以上比较,用best.
第一句有三个对象比较（红的,黄的,蓝的）,第二句有三个以上对象比较（她和班级其他人）,根据上述规律,建议都用best.

第一句，which is the best更规范一点，口语中没那么讲究，国外网页用which is best的也大有人在。第二句，只能用best。如果硬要加the，可以这样说：She is the best friend of mine in the class. 语法细节上不专业，凭经验作答，仅供参考。
